Lysine Post-Translational Modifications

Among the 20 amino acid residues in proteins, lysine (K) is one of the most common targets for various post-translational modifications (PTMs). With the rapid development of high-resolution mass spectrometry (MS) and immunoprecipitation purification techniques, multiple acylation reactions on lysine have been discovered in the past decade, including acetylation, succinylation, malonylation, glutarylation, crotonylation, butyrylation, and lactylation. Reversible lysine post-translational modifications (Lys-PTMs) are ubiquitous biochemical processes that play indispensable roles in regulating numerous key cellular functions. Due to their close association with gene transcription, cellular metabolism, aging, and various diseases, lysine modifications have been a hot and focal point of scientific research.

Using the MalAM-yne probe for metabolic labeling at the cellular level, followed by click chemistry reactions at the lysate level, magnetic bead enrichment, and enzymatic digestion, specific identification of proteins with malonylation modifications is achieved through high-resolution mass spectrometry.
